Junior guard Zahna Medley led TCU (18-14) with a game-high 26 points, going 6-of-12 from 3-point range and a perfect 6-for-6 from the charity stripe.
Senior forward Chelsea Prince ended her career with her fourth double-double, posting 13 points and 11 rebounds while adding four assists and four steals.
Senior guard Donielle Breaux also scored in double figures with 13 points to go with eight boards, while fellow senior Natalie Ventress ended her storied career in the Purple and White with six points and nine rebounds.
After holding a two-point halftime lead, the Horned Frogs controlled much of the second half before Southern Miss went on a 14-2 run over the final 2:50 of regulation to force overtime.
Medley gave TCU its first double-double digit lead with a pair of 3-pointers early in the second half and Ventress followed with an offensive rebound and 3-point play to put TCU ahead 38-26 at the 14:49 mark.
Medley buried her fifth trey of the contest with 5:47 remaining to give TCU its largest lead of the contest at 53-39.
The Golden Eagles' Jerontay Clemons then took over, as the athletic guard made several drives to the basket, scoring 12 of Southern Miss' next 14 points.
With TCU clinging to a five-point lead with 1:04 remaining, Clemons converted a layup and was fouled to trim the advantage to three points.
Clemons then rebounded her own miss and TCU was whistled for another foul, where she converted both shots from the charity stripe to make it a 61-60 contest with 42.9 seconds left.
Clemons split a pair of free-throws with 17.8 seconds left to tie the game, and the Frogs were able to get a stop on the Golden Eagles' final possession to force the overtime period.
Medley drained her sixth long-range bucket to even the score at 64-64 with 3:08 remaining, but USM's Brooke Rhodes answered with back-to-back trifectas to put the contest away.
TCU made its second straight trip to the postseason this season while earning its first postseason victory since 2008 on Thursday against Stephen F. Austin.
